چکیده

Human platelet concentrates stored at 22#{176}C for transfusion purposes progressively lose their in vitro sensitivity to single aggregating agents such as adenosine diphosphate (ADP). It is known that very small amounts of epinephrine. collagen. or arachidonic acid can markedly enhance the aggregation of platelets in fresh platelet-rich plasmas in response to concentrations of ADP that do not cause aggregation when used singly. Therefore. we investigated the effects of pairs of aggregating agents on eight platelet concentrates on the day of preparation and after 3 and 5 days of storage at 22#{176}C. We studied platelet aggregation and secretion with a lumi-aggregometer and thromboxaneB2 formation using a radioimmunoassay. When aggregation was tested in response to single agents. stored platelets were completely unresponsive to epinephrine. markedly insensitive to ADP, and moderately insensitive to collagen and arachidonic acid. In spite of this. there was no statistically significant loss of sensitivity to aggregating agents

Ultrastructural changes in human platelets during arachidonic acid-induced aggregation.

This study was undertaken to visualize the sequence of ultrastructural changes occurring in human platelets during aggregation induced by arachidonic acid. Aggregometer recordings were obtained up to the time of adding glutaraldehyde to begin fixation of platelets for electron microscopy. A correlation was shown between the degree of aggregation and changes in the fine structure of human platel...

Monitoring the entry of new platelets into the circulation after ingestion of aspirin.

There have been reports of a 24-48-hr delay in the recovery of platelet cyclooxygenase activity and platelet function after the ingestion of aspirin. However, these studies employed a single aggregating agent to stimulate enzymatic or functional activity. We investigated the effects of some pairs of aggregating agents on 14 platelet-rich plasmas (PRP) from normal subjects 2 and 4 hr after inges...

Platelet aggregation in whole blood from patients with Glanzmann's thrombasthenia.

We examined platelet aggregation in platelet-rich plasma (PRP) and in whole blood from two patients with Glanzmann's thrombasthenia. In PRP, aggregation was measured by monitoring the changes in light absorbance that occurred in response to aggregating agents; to measure platelet aggregation in whole blood, we used a platelet counting technique. In PRP, the patients' platelets showed defective ...

The membrane potential of human platelets.

The membrane potential of the human platelet was investigated using the membrane potential probes 3,3'-dipropyl-2,2'-thiadicarbocyanine iodide and tritiated triphenylmethylphosphonium bromide. The membrane potential in physiologic buffer was estimated to be 52-60 mV inside negative. The membrane was depolarized when extracellular potassium or hydrogen ion concentrations were increased. Changes ...

Inhibition of platelet aggregation by protease inhibitors. Possible involvement of proteases in platelet aggregation.

The possible participation of proteases in human platelet aggregation was explored using various protease inhibitors and substrates. Protease inhibitors used included naturally occurring inhibitors of serine proteases and synthetic inhibitors that modify the active site of protease. Substrates used were synthetic substrates for the trypsin type as well as for the chymotrypsin type of protease. ...
